Production of a Plasma Derived Universal Antivenom against All Elapid Neurotoxic Snake Venoms

چکیده

Snakebite envenoming has killed about 138000 people and maimed 400,000 victims annually. WHO designated this medical problem as one of the most neglected tropical diseases for which effective, affordable antivenoms (AVs) are urgently needed. Production potent AV against neurotoxic venoms was difficult thought to be due low immunogenicity postsynaptic neurotoxins (PSNT) cause death in victims. However, it showed that use ineffective adjuvant immunization horse root cause. The highly effective Freund (FA) causes severe local reactions could not used. A novel protocol termed ‘low dose volume multi-site’ tested shown obliterate side effect allow safe FA horse. led production 7 monovalent AVs, 2 polyvalent 4 another 3 hematotoxic venoms. These AVs treatment snakebite without need identify culprit snakes. Furthermore, we have a strategy using ‘Diverse toxin repertoire’ 12 Asian elapid fractions. resulting antiserum effectively neutralized at least 36 28 species encompassing 10 genera from 20 countries on continents, likely all snake results indicate universal antivenom world can produced save numerous lives.

New techniques in antivenom production and active immunization against snake venoms.

Standard methods of commercial antivenom production are costly both in terms of animals, their maintenance, the use of large amounts of expensive venoms and man-hours. The basic technology, which involves the hyperimmunization of horses, bleeding and subsequent purification of the plasma immunoglobulin G (IgG) using salt precipitation and pepsin digestion, has not advanced significantly for man...

Molecular cloning of serine proteases from elapid snake venoms.

Serine proteases are widely distributed in viperid snake venoms, but rare in elapid snake venoms. Previously, we have identified a fibrinogenolytic enzyme termed OhS1 from the venom of Ophiophagus hannah. The results indicated that OhS1 might be a serine protease, but there was no structural evidence previously. In the present study, the primary structure of OhS1 was determined by protein seque...

Cross Neutralization of Afro-Asian Cobra and Asian Krait Venoms by a Thai Polyvalent Snake Antivenom (Neuro Polyvalent Snake Antivenom)

BACKGROUND Snake envenomation is a serious public health threat in the rural areas of Asian and African countries. To date, the only proven treatment for snake envenomation is antivenom therapy. Cross-neutralization of heterologous venoms by antivenom raised against venoms of closely related species has been reported. The present study examined the cross neutralizing potential of a newly develo...

Proteolytic activity of Elapid and Viperid Snake venoms and its implication to digestion

Testing whether venoms may aid in digestion of the prey, eleven snake venoms were compared for the presence of proteases and endopeptidases that function in alkaline pH conditions. In vitro experiments examined the relative protease and endopeptidase activity of the venoms, which involved combining bovine muscle and snake venom in a buffered solution, encased within dialysis tubing. This mixtur...

Comparison of the in vitro neuromuscular activity of venom from three Australian snakes (Hoplocephalus stephensi, Austrelaps superbus and Notechis scutatus): efficacy of tiger snake antivenom.

1. Tiger snake antivenom, raised against Notechis scutatus venom, is indicated not only for the treatment of envenomation by this snake, but also that of the copperhead (Austrelaps superbus) and Stephen's banded snake (Hoplocephalus stephensi). The present study compared the neuromuscular pharmacology of venom from these snakes and the in vitro efficacy of tiger snake antivenom. 2. In chick biv...
